The third isolate had a novel allele at the
abcZ locus and was assigned ST-4242.
MLST was performed by determining the nucleotide sequences of 7 housekeeping genes (
abcZ, adk, aroE, fumC, gdh, pdhC, pgm) as previously described (8).
MLST was performed by using the following seven housekeeping genes (protein products are shown in parentheses):
abcZ (putative ATP-binding cassette transporter), adk (adenylate kinase), aroE (shikimate dehydrogenase), fumC (fumarate hydratase) gdh (glucose-6-phosphate dehydrogenase), pdhC (pyruvate dehydrogenase subunit), and pgm (phosphoglucomutase) as previously described (6).
PFGE was done by digesting chromosomal DNA prepared in agarose blocks with NheI and SpeI as described (22), and MLST by sequencing gene fragments of
abcZ, adk, aroE, fumC, gdh, pdhC, and pgm, also as described (16; http://www/ mist.net).
For MLST, the primers of the housekeeping genes
abcZ (putative ABC transporter), adk (adenylate kinase), aroE (shikimate dehydrogenase), gdh (glucose-6-phosphate dehydrogenase), pdhC (pyruvate dehydrogenase subunit), and pgm (phosphoglucomutase) were synthesized by our institute according to the sequences published by Maiden et al.
